| I. Background | |
| CD46 基因编码的膜辅蛋白（MCP）是一种广泛表达于所有有核细胞的跨膜糖蛋白， 属于补体调节蛋白家族，在免疫稳态和病原体防御中扮演关键角色。其核心功能包括：作 为补体系统的重要调节因子，顺利获得辅助因子 I 裂解 C3b 和 C4b，保护宿主细胞免受补体过 度激活导致的损伤；同时作为 T 细胞共刺激分子，诱导调节性 T 细胞（Tregs）分泌抗炎因 子 IL-10，抑制过度免疫反应，维持免疫平衡。 CD46 功能异常与多种疾病密切相关。基因突变可导致非典型溶血性尿毒症综合征 （aHUS），表现为微血管溶血性贫血、血小板减少和急性肾衰竭；在自身免疫性疾病中， 多发性硬化（MS）患者因 CD46 信号缺陷导致 IL-10 分泌不足，加剧神经炎症；类风湿关 节炎患者则因 CD46 功能异常，T 细胞无法有效抑制促炎因子。此外，CD46 在肿瘤组织中 高表达，参与免疫逃逸机制，成为癌症治疗的新靶点。 |
