| I. Background | |
| ITGA6 与 ITGB4 是整合素家族的关键成员，分别编码α6 和β4 亚基，顺利获得非共价结合 形成α6β4 整合素复合体，在细胞粘附、迁移及信号传导中发挥核心作用。ITGA6 位于 2q31.1，编码整合素α6 亚基，可与β1 或β4 结合，参与细胞与层粘连蛋白的相互作用，调控 细胞增殖、组织再生及肿瘤侵袭，其异常表达与交界性大疱性表皮松解症、卵巢癌、结直 肠癌及肾病综合征相关，是潜在治疗靶点。ITGB4 位于 17q25，编码整合素β4 亚基，与α6 结合后形成层粘连蛋白受体，顺利获得 FAK/Src、EGFR-AKT 等信号通路影响细胞极性、肿瘤 进展及耐药机制，其突变直接导致伴幽门闭锁的大疱性表皮松解症，并在胰腺癌、非小细 胞肺癌中高表达，与患者预后密切相关。两者形成的α6β4 整合素在肿瘤生物学中尤为关键， 如在卵巢癌中表达显著上调且呈正相关，可作为诊断标志物；在胰腺癌中则顺利获得调控 AREG、EREG 和 MMP1 等基因促进侵袭转移。 | |
